Understanding Single Chain Variable Fragments

Single-chain variable fragments (scFvs) are essential building blocks for breakthrough therapeutics including bispecific antibodies and CAR-T cell therapies. They are composed of the variable heavy (VH) and variable light (VL) domains joined by a flexible peptide linker.

scFvs retain antigen-binding activity, are straightforward to engineer, and can be readily fused to other proteins. Owing to their small size, they often show improved tissue penetration when used alone, although they also have a shorter half-life.

However, the absence of constant domains increases their tendency to aggregate and misfold, which can make bispecific formats containing scFvs less stable – leading to:

  • High development costs
  • Poor therapeutic performance
  • Manufacturing challenges and reduced yields
  • Project cancellations

For three decades, the field has lacked a broadly applicable solution to the scFv stability problem. Until now.

Optimized CHO Expression for Challenging Antibody Formats

evitria now offers Opti-mAb® technology from AAX Biotech – a proprietary molecular stabilization strategy for scFvs.

This significantly improves stability of scFvs and scFv-containing bispecific antibodies.
The result: higher titers, improved quality, and consistent manufacturability for even the most complex antibody constructs.

“Opti-mAb® represents a significant advancement in scFv development. By combining AAX Biotech’s proprietary optimization with our 15 years of exclusive CHO expertise, we can now deliver exceptional results for constructs that previously challenged conventional platforms.”

Opti-mAb®: A Molecular Stabilization Breakthrough

AAX Biotech’s Opti-mAb® technology provides the first generally applicable molecular stabilization strategy for scFv antibodies.

Now available exclusively through evitria’s CHO expression platform, Opti-mAb® delivers:

  • Increased Production Yields: Significantly higher titers compared to unstabilized scFv constructs.
  • Eliminated Aggregation: Molecular stabilization prevents the aggregation issues that have historically limited scFv applications.
  • Enhanced Therapeutic Properties: Improved stability translates directly to better performance in bispecific antibodies and CAR-T therapies.
  • Accelerated Development: Overcome the stability bottleneck that has delayed or canceled promising projects for decades.

FAQs about Opti-mAb® Technology

Opti-mAb® typically delivers 2-40x higher expression titers compared to non-optimized scFvs. This makes it particularly valuable for challenging constructs that yield poorly in standard platforms or require large quantities for in vivo studies.

Yes. Opti-mAb® acts at the format design stage and creates optimized scFv sequences that are fully compatible with industry-standard expression systems, such as CHO cells. This ensures that the candidates identified during discovery can be seamlessly transitioned to clinical-scale manufacturing without requiring further re-engineering or facing comparability issues.

Opti-mAb® can be used for any scFvs- based formats, including complex bispecifics in any orientation, fusion proteins and stand-alone scFvs.

Yes. Expedited timelines are available for urgent projects. Contact our team to discuss your specific timeline requirements and we’ll design a production strategy to meet your milestones.

Absolutely. Unlike providers with rigid packages, we individually discuss and design analytical strategies to match your specific needs – from basic QC to comprehensive characterization including mass spectrometry, glycan profiling, and functional assays.
