Understanding Single Chain Variable Fragments
Single-chain variable fragments (scFvs) are essential building blocks for breakthrough therapeutics including bispecific antibodies and CAR-T cell therapies. They are composed of the variable heavy (VH) and variable light (VL) domains joined by a flexible peptide linker.
scFvs retain antigen-binding activity, are straightforward to engineer, and can be readily fused to other proteins. Owing to their small size, they often show improved tissue penetration when used alone, although they also have a shorter half-life.
However, the absence of constant domains increases their tendency to aggregate and misfold, which can make bispecific formats containing scFvs less stable – leading to:

- Poor therapeutic performance
- Manufacturing challenges and reduced yields
